Gene Synthesis> | The DA used in this study was purchased from Sigma Chemical Company, St. Louis, MO. The two GnRH isoforms used in this study belong to a lower vertebrate (lamprey) and invertebrate (octopus). The lGnRH-III isoform is recognized being within the GnRH-IV group (Decatur et al., 2013, Zhang et al., 2008), whereas an octGnRH is recognized as GnRH-V (Kornthong et al., 2013, Roch et al., 2011). The GnRH peptides, octGnRH (pGlu-Asn-Tyr-His-Phe-Ser-Asn-Gly-Trp-His-Pro-Gly-NH2) and lGnRH-III (pGlu-His-Trp-His-Asp-Trp-Lys-Pro-Gly-NH2), were custom synthesized (GenScript, Piscataway, NJ, USA). The neurotransmitter and neuropeptides were dissolved in 0.9% normal saline. An antibody to octGnRH was produced previously and its specificity tested rigorously (Tinikul et al., 2011a). In addition, a polyclonal antibody against lGnRH-III was produced previously and its specificity was also tested rigorously (Saetan et al., 2013). The female New Zealand white rabbits (8 week-old) were obtained from the Animal Care Unit, Faculty of Science, Mahidol University, and used for production of anti-GnRHs with the approval of the Mahidol Animal Ethics Committee. We investigated the effects of two isoforms of gonadotropin-releasing hormone (GnRH), namely octopus GnRH (octGnRH) and lamprey GnRH-III (lGnRH-III), and a neurotransmitter, dopamine (DA), on ovarian maturation and spawning in the Pacific white shrimp, Litopenaeus vannamei. The effects of GnRHs and DA were examined by evaluating gonadosomatic indices (GSIs), oocyte diameters (ODs), proliferation of the germ cells, and hemolymph vitellogenin (Vg) concentrations.
