First Report of Downy Mildew Caused by Hyaloperonospora sisymbrii-sophiae on Flixweed (Descurainia sophia) in China

DNA was extracted from infected leaves with the onset of fungal hyphae, sporangiophore, and sporangia, and extracts tested by PCR using primers targeting conserved regions of cytochrome oxidase (COX2) (Choi et al. 2015; Hudspeth et al. 2000) and the internal transcribed spacer (ITS) region of rDNA (White et al. 1990). PCR products of 885 and 631 bp for the rDNA ITS region and mtDNA COX2 regions were obtained, respectively. These were sequenced in GenScript (Nanjing) Co., Ltd., deposited in GenBank (accession nos. KC414198 and KY417137), and compared with published sequences. These sequences were found to be 99% identical. A maximum phylogenetic analysis of the ITS sequences (855 bp) confirmed that the symptoms observed on flixweed in China were caused by H. sisymbrii-sophiae. These morphological observations and molecular evidence showed that the downy mildew symptoms observed in flixweed were caused by H. sisymbrii-sophiae and recorded for the first time in China. |
